Bacterial Leaf Blight (BLB) of rice, a great concern to the rice growing countries especially in Bangladesh where the rice is cultivated as a staple food crop. Development of resistant cultivar is the best approach to control the disease.
Mubassir et al. (2016) carried out an intensive study jointly by the Department of Biotechnology, Bangladesh Agricultural University, Department of Plant Pathology, Bangladesh Agricultural University and Bangladesh Institute of Nuclear Agriculture (BINA) with a view to exploring suitable lines/varieties resistant to the disease through using morphological and molecular screening techniques. The test materials such as ten advanced lines and seventeen varieties of rice including the check resistant and susceptible cultivars were collected from Bangladesh Institute of Nuclear Agriculture, Bangladesh Rice Research Institute and Gazipur district of Bangladesh.

It may be mentioned that, finding out a suitable line for developing as a commercial variety is always challenging and a rice line can only be suggested for developing as commercial variety if it fulfils the criteria of those characteristics for what it is being developed. Scientists used to develop the best BLB resistant cultivar through continuous endeavour of morpho-molecular screening as the initial steps. The morphological screening was done after 21 days of inoculation by an active bacterial strain. The bacterial strain of Xanthomonas oryzae pv. oryzae (Xoo.) “BXO-09” was used for the inoculation experiments which is the best active strain of Xoo. in Bangladesh. The inoculation was done following clipping method. Simultaneously, to extract genomic DNA, Juvenile, fresh green leaf samples were collected from 21-25 days old seedlings and for molecular study DNA amplification was done by different RFLP markers.
The detail reports on the study have already been published in a reputed journal “Journal of Plant Sciences”. Several rice lines were found moderately resistant to BLB. Also the article illustrated that the rice lines and varieties which were found as resistant, with similar kind of banding pattern in molecular study of DNA amplification by three Restriction Fragment Length Polymorphism (RFLP) molecular markers RG136, RG556 and pTA248.
“After doing morphological and molecular analysis for bacterial blight resistance, it has been found some promising rice lines which can be developed as commercial variety upon further study”, said M H M Mubassir. The authors believe that the findings may be successfully incorporated by the breeders in their relevant as well as the concerned breeding program.
